I recently picked up an old Warmoth Strat neck used for cheap.
It is made/drilled for the vintage style Klusons.
I have a set of locking tuners that are a larger diameter post size, so I need to enlarge the holes in the headstock.

I have watched a lot of Youtube videos, and found this one.
It is short, to the point, simple and best of all seems to work. (I have tried it on a block of practice wood.)

He used a 10 mm drill bit to reference the reamer against.
I do not have a 10 mm drill bit, but I have a 3/8, which is slightly smaller.
I just kept going with the reamer, checking frequently, until the post hole was the right size.

As for the screw/anchor pin holes, I have a pin vise that I used on a friends' guitar to do them manually.
It's slower than a power drill, but I'd rather not take any chances with a power tool.
